Perhaps you have this error if you have move the environment from one server to another one.
The error message tell you that the file "index.php" located in the directory D:\xampp\htdocs\Multi-site\multisites\site\ is not able to find a file "defines.php" that is located in the D:\xampp\htdocs\Multi-site\multisites\site\includes

So you have to check if this file exists or check the permission.

An additional question is do you have the symbolic link available and have you deployed the slave site in to a specific directory.
Something "strange" is the you seems to use a Windows platform with "Xamp" that seems to be unix.

So check that you have the path expected.
